Toulouse's Kinghorn has recovered from the injury that ruled him out of rounds one and two of the Championship to take his place at fullback.
The 27-year-old mostly deputised for Stuart Hogg at 15 in last year's Guinness Men's SIx Nations, before bursting to prominence at fly-half in a hat-trick performance in the closing round against Italy.
Following an agonising loss to France in round two, Scotland are now setting their sights on a fifth win in seven Calcutta Cup meetings with England. Victory in Edinburgh would equal their longest winning streak of four matches in this fixture, which was achieved between 1893-1896.
